COX6a2 Inhibitors

Antimycin A, Rotenone, and Oligomycin act on different complexes of the electron transport chain but ultimately reduce the efficiency of electron transfer to the COX complex. Compounds such as Cyanide, Azide, Carbon monoxide, Hydrogen sulfide, and Sodium azide directly target the COX complex, specifically the cytochrome c oxidase activity, thereby impeding the function of COX6a2. Their inhibition is due to their high affinity for the heme iron within the enzyme, which is essential for its catalytic activity. By obstructing the terminal step in the electron transport chain, these inhibitors significantly impact cellular respiration.

Zinc and Penicillamine interact with the metal centers that are critical for the structural integrity and function of enzyme complexes, including the COX complex. Zinc's inhibitory effects arise from its potential to displace necessary metal ions, while copper chelators sequester copper, depriving COX6a2 of its requisite cofactor.
