Similarly, IBMX, by inhibiting phosphodiesterases, maintains high levels of cAMP, supporting the activation of PKA and subsequent phosphorylation of CGI-121. On another front, PMA and TPA, both activators of protein kinase C (PKC), initiate a signaling pathway that leads to the phosphorylation of specific target proteins, potentially including CGI-121, thereby fostering its activation.
Further, the increase of intracellular calcium levels through the action of ionophores like Ionomycin and A23187 can activate calmodulin-dependent kinases, which also have the capacity to phosphorylate and activate CGI-121. Thapsigargin, by inhibiting the SERCA pump, leads to a similar rise in intracellular calcium, potentially triggering the activation of CGI-121 through calcium-sensitive kinases. Okadaic Acid and Calyculin A, both inhibitors of protein phosphatases PP1 and PP2A, prevent the dephosphorylation of proteins, thereby maintaining CGI-121 in an activated phosphorylated state. Anisomycin, while primarily known as a protein synthesis inhibitor, activates stress-activated protein kinases, which could result in the phosphorylation and subsequent activation of CGI-121. Finally, EGF can activate the MAPK/ERK pathway, which is known to phosphorylate a variety of proteins, possibly including CGI-121, leading to its activation, while Bt2cAMP, a cAMP analog, directly activates PKA, which then can phosphorylate and activate CGI-121.
