CES3 Activators

The CES3 Activators are a chemical class that modulates the CES3 enzyme, a critical player in fatty acid metabolism and drug metabolism. These activators operate through various cellular mechanisms that can lead to increased CES3 activity. 4-Methylumbelliferone and Indomethacin function by decreasing the levels of molecules that normally inhibit CES3 activity. 4-Methylumbelliferone reduces hyaluronic acid synthesis, which indirectly enhances CES3 activity, given that higher hyaluronic acid levels typically suppress CES3 function. Indomethacin, as a COX inhibitor, curtails the production of prostaglandins, which normally dampen CES3 activity.

Another key group of CES3 activators is the PPAR agonists, which include Troglitazone, Fenofibrate, Rosiglitazone, Pioglitazone, GW7647, GW1929, and WY-14643. These compounds, by binding to and activating PPARα or PPARγ, can boost CES3 expression, thereby enhancing its activity. PPARs are nuclear hormone receptors that regulate gene expression, and their activation leads to increased CES3 transcription. Dexamethasone, a glucocorticoid receptor agonist, also enhances CES3 activity by elevating its expression. The glucocorticoid receptor, when activated, can upregulate a variety of genes, including CES3. Finally, Metformin and AICAR activate AMPK, which can indirectly lead to CES3 activation. AMPK activation spurs upregulation of PPARα, a known CES3 activator.
