Oxidative stress responsive serine-rich 1 (OSR1) is a protein encoded by the OSR1 gene in humans. This protein is implicated in the regulation of ion transport and homeostasis, particularly involving the modulation of sodium (Na+) and potassium (K+) ion channels and transporters. OSR1 belongs to the STE20/SPS1-related proline/alanine-rich kinase (SPAK) subfamily of serine/threonine protein kinases, and it is activated in response to cellular stress, including hyperosmotic and oxidative stress.

At the molecular level, the activation of OSR1 is closely tied to the upstream kinases of the With No Lysine (WNK) signaling pathway. These kinases phosphorylate and activate OSR1, which in turn phosphorylates and modulates the activity of various cation-Cl- cotransporters (CCC), including the Na-K-2Cl cotransporter (NKCC) and the Na-Cl cotransporter (NCC). This modulation is crucial in the control of ion balance across cellular membranes, which is vital for maintaining cell volume and the osmotic balance necessary for cell survival and function.OSR1 plays a significant role in the physiological regulation of blood pressure and osmotic stability by influencing kidney function. Therefore, dysregulation of OSR1 activity has been associated with hypertension and other cardiovascular disorders. The protein is also part of a broader network of signaling pathways that respond to changes in the cellular environment, highlighting its importance in cellular adaptation mechanisms.
