Forskolin and IBMX focus on elevating cellular cAMP levels, thereby activating Protein Kinase A (PKA). PKA can phosphorylate C/EBP α, which enhances its transcriptional activity. Rosiglitazone, a PPARγ agonist, activates C/EBP α through a cross-talk mechanism between the PPARγ and C/EBP α pathways. 8-Br-cAMP, a synthetic analog of cAMP, activates PKA and subsequently influences C/EBP α activation. MG132 is a proteasome inhibitor that sustains C/EBP α levels by preventing its degradation. Similarly, LY294002 inhibits PI3K to modulate C/EBP α by blocking its negative regulatory pathways.
Another subset of chemicals acts indirectly but effectively on C/EBP α.
