Chemical activators of fibronectin type III domain containing 10 (FNDC10) include a variety of compounds that initiate intracellular signaling cascades resulting in the protein's activation. Phorbol 12-myristate 13-acetate, commonly known as PMA, is a potent activator of protein kinase C (PKC). PKC plays a crucial role in phosphorylating FNDC10, thereby leading to its functional activation. An analogous effect is observed with hydrogen peroxide, which activates PKC, mitogen-activated protein kinases (MAPKs), and phosphoinositide 3-kinases (PI3K), all of which are capable of targeting and activating FNDC10 through phosphorylation. Forskolin, through the elevation of cyclic AMP (cAMP) levels, activates protein kinase A (PKA), which then can target FNDC10 for phosphorylation, facilitating its activation. Dibutyryl-cAMP, a cAMP analog, mimics this activation route by directly stimulating PKA, which in turn activates FNDC10.
Calcium ionophores like A23187 and Ionomycin increase intracellular calcium levels, which activate calcium-dependent protein kinases, leading to the phosphorylation and activation of FNDC10. Similar calcium-mediated activation can be indirectly influenced by Zinc Chloride, as zinc ions are cofactors for numerous kinases that can phosphorylate FNDC10. Epidermal Growth Factor (EGF) binds to its receptor, triggering a downstream signaling cascade involving MAPKs that activate FNDC10. Insulin also promotes the activation of FNDC10 through the PI3K/Akt signaling pathway, which has been implicated in the phosphorylation and subsequent activation of various proteins, including FNDC10. Inhibition of dephosphorylation processes is another strategy to maintain FNDC10 in an active state, as demonstrated by Okadaic Acid, an inhibitor of protein phosphatases 1 and 2A, which indirectly maintains FNDC10 in a phosphorylated, active form. Sodium Fluoride activates G-proteins that stimulate MAPK pathways, leading to the activation of FNDC10. Lastly, Anisomycin is a powerful activator of MAPKs, which phosphorylate FNDC10, resulting in its activation. Through these diverse but interconnected biochemical pathways, FNDC10 is functionally activated by these chemical compounds.
