ADAM10 Activators

ADAM10 activators are compounds that stimulate the activity or expression of ADAM10 (A Disintegrin and Metalloprotease 10). ADAM10 is a member of the ADAM protein family, a group of transmembrane and secreted proteins with a wide range of functions, including cell adhesion, migration, and proteolytic processing. As a zinc-dependent metalloprotease, ADAM10 is involved in the "shedding" process, where it cleaves the extracellular domains of various cell surface proteins, releasing them from the cell membrane. This ectodomain shedding is a critical post-translational modification that regulates the availability and functionality of many membrane-associated proteins.

ADAM10 is recognized for its role in the cleavage of numerous substrates, including growth factors, cytokines, receptors, and adhesion molecules. One of its most extensively studied substrates is the amyloid precursor protein (APP). Cleavage of APP by ADAM10 is non-amyloidogenic, meaning it prevents the formation of amyloid-beta peptides, which are associated with neurodegenerative conditions. Additionally, ADAM10 is involved in the processing of Notch, a critical signaling molecule in cell differentiation and development. Activation of ADAM10 can influence these and other pathways by modulating the availability of functional proteins. ADAM10 activators, by enhancing the enzyme's activity or expression, can influence various biological processes where ADAM10-mediated shedding plays a role.
