Chemical activators of double homeobox family member 3 can engage various intracellular signaling pathways to induce the protein's activation. Forskolin, a potent activator of adenylate cyclase, increases the intracellular levels of cyclic AMP (cAMP), which in turn activates protein kinase A (PKA). The activated PKA can then phosphorylate target proteins, including double homeobox family member 3, enhancing its activity. Similarly, IBMX, by inhibiting phosphodiesterases, prevents the breakdown of cAMP, thereby indirectly maintaining PKA in an active state which can lead to the phosphorylation and consequent activation of double homeobox family member 3. Anisomycin, although primarily known as a protein synthesis inhibitor, can activate stress-activated protein kinases that may phosphorylate and activate double homeobox family member 3 as part of a cellular stress response.

In a parallel pathway, the protein kinase C (PKC) activators such as PMA and Bryostatin 1 can lead to the phosphorylation and activation of double homeobox family member 3. PKC, when activated by these compounds, can modify a variety of substrates, including double homeobox family member 3. Calcium plays a vital role in the activity of certain protein kinases, and ionophores like Ionomycin and A23187 elevate intracellular calcium levels, which can activate calcium-dependent kinases that subsequently phosphorylate and activate double homeobox family member 3. Thapsigargin, by inhibiting the SERCA pump, leads to a similar increase in cytosolic calcium, again potentially resulting in the activation of double homeobox family member 3 through kinase-mediated phosphorylation. Furthermore, inhibitors of protein phosphatases such as Okadaic Acid and Calyculin A prevent the dephosphorylation of proteins, leading to a net increase in phosphorylation and activation of proteins, including double homeobox family member 3, by maintaining them in a phosphorylated state.
