Topo IIβ Antibody (H-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Topo II beta protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Topo II beta antibody (H-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Topo II beta protein plays a crucial role in DNA replication and transcription by introducing transient double-strand breaks in DNA, allowing for relaxation of supercoiled DNA, which is essential for proper chromosomal segregation during cell division. This function is particularly important in rapidly dividing cells, where DNA topology regulation is vital for maintaining genomic stability. Differential expression of Topo II isoforms, including Topo IIα and Topo IIβ, suggests distinct roles in cellular processes, with Topo IIβ being implicated in specific tissue functions and responses to DNA damage. Understanding Topo IIβ′s unique contributions can provide insights into potential therapeutic targeting in cancer treatment, where topoisomerase inhibitors are commonly used to disrupt cancer cell proliferation.

I am using Topo IIβ (H-8): sc-25330 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 1 out of 5 by from TOPOIIB H-8 in Western BlotI received this as a free sample antibody and tested it out at a 1:200 dilution using Whole Cell Extracts from four different cell lines (HeLa, HEK293, HEK293T, and Mouse Embryonic Fibroblast cells). The expected size for TOPO11B is 180 kDa but I didn't see anything in any of the lanes. The exposure of the blot was weird for this antibody and another Topoisomerase antibody I also tested (Santa Cruz A-12, though this one at least had bands at the correct size).

Rated 5 out of 5 by from a good antibody for IFThis antibody stains the nucleus with a stronger signal in the nucleoli (1:200, formaldehyde-fixed HeLa). However, we did not have much luck with western analysis using U251 cell extract (1:500 dilution).
