Smad6 Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Smad6 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). anti-Smad6 antibody (D-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Smad6 protein plays a crucial role in regulating TGFβ/BMP signaling pathways, acting as a negative regulator that inhibits phosphorylation of other Smad proteins, thereby modulating cellular responses to growth factors. This function helps maintain cellular homeostasis and prevents excessive signaling that could lead to pathological conditions such as fibrosis or cancer. Through interference with Smad2 and Smad3 activation, Smad6 ensures a balanced response to TGFβ and activin, which are vital for various developmental processes and tissue homeostasis. We recently order Smad6 Antibody (D-4) X for ChIP and it did not work in our case. I would like to know if there is a paper where this antibody was used for ChIP applications? Maybe this can help, because until the moment I could not find any...

When should the TransCruz reagent sc-25321 X be used instead of the standard form sc-25321?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
