Sigma Receptor Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects Sigma Receptor of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Sigma Receptor (B-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Sigma Receptor, also known as opioid receptor sigma 1 (Oprs 1), plays a crucial role in modulating intracellular signaling pathways, particularly in response to stress and pain. Sigma Receptor is primarily located in the endoplasmic reticulum and plasma membrane of neurons, where Sigma Receptor influences calcium signaling and neurotransmitter release, making Sigma Receptor vital for processes such as analgesia and neuroprotection. Sigma Receptor localization allows for rapid responses to various stimuli, including modulation of opioid effects and regulation of neurochemical systems involved in mood and cognition. Sigma Receptor is implicated in several psychiatric disorders and central nervous system diseases, highlighting Sigma Receptor importance in both health and disease. Pregnenolone and dehydroepiandrosterone, which act as Sigma Receptor agonists, further illustrate Sigma Receptor′s role in neurobiology, while progesterone serves as a potent antagonist, showcasing the complexity of these interactions. Hello, I am looking to use immunofluorescence to view this protein in cancer cells. Is there a recommended permeabilization technique for this membrane embedded receptor (saponin/triton x100 etc)?

Thank you for your question. We have not tested this mouse monoclonal, Sigma Receptor Antibody (B-5): sc-137075, by immunofluorescence in cancer cells and have not determined the optimal permeablization technique for this application. We have tested the antibody in formalin-fixed paraffin-embedded tissue sections and recommend using sodium citrate buffer (pH 6) and heat treatment for antigen retrieval. We would recommend consulting some of the product citations on our website to see how other researchers have used this antibody for IF, e.g., PMID: 24803543.

We recommend using conjugated primary antibodies. This product, sc-137075, is available directly conjugated to FITC, AF488, and AF 647.
