    PTEN Antibody (H-3) is a mouse monoclonal IgG2b kappa light chain antibody specifically designed to recognize the N-terminal epitope of PTEN protein, also known as phosphatase and tensin homolog, MMAC1, or TEP1. Anti-PTEN antibody (H-3) targets amino acids 2-28 of human PTEN and is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) across a wide range of species, including mouse, rat, human, equine, canine, bovine, porcine, and avian origins. PTEN is a crucial tumor suppressor protein primarily localized in both cytoplasm and nucleus of cells. In cytoplasm, PTEN regulates cell growth, proliferation, and survival by dephosphorylating phosphatidylinositol (3,4,5)-trisphosphate, thereby antagonizing the PI3K/AKT signaling pathway, which is essential for preventing uncontrolled cell division. Nuclear presence is equally important, as PTEN contributes to maintaining genomic stability and regulating DNA repair processes, which are vital for preventing mutations that can lead to cancer. Dual localization of PTEN allows tumor-suppressive functions to perform effectively, ensuring cellular processes remain balanced and potential malignant transformations are inhibited. Loss or mutation of PTEN disrupts these critical regulatory mechanisms, often resulting in development and progression of various cancers, including glioblastomas, breast, kidney, and prostate cancers. PTEN monoclonal antibody (H-3) is recommended for researchers studying distribution and functional dynamics of PTEN within different cellular compartments, cellular homeostasis, and preventing tumorigenesis.
