PERK Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 21-320 mapping near the N-terminus of human PERK and reacts with proteins from mouse, rat, and human origins; anti-PERK antibody (B-5) is suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry. PERK, also known as eukaryotic translation initiation factor 2-alpha kinase 3 (EIF2AK3), is a type I transmembrane protein located in the endoplasmic reticulum (ER) that plays a critical role in cellular stress responses; PERK′s primary function is to detect the accumulation of misfolded proteins within the ER lumen and initiate the unfolded protein response (UPR) by phosphorylating the alpha subunit of eukaryotic initiation factor 2 (eIF2α). This phosphorylation event reduces general protein synthesis, thereby decreasing the influx of new proteins into the ER and allowing the cell to restore normal function, while selectively upregulating stress-related genes such as ATF4 that promote adaptation and survival under stress conditions. Understanding PERK′s function is important because dysregulation of the UPR and ER stress responses are implicated in various diseases, including neurodegenerative disorders, diabetes, and cancer; studying PERK can provide valuable insights into the mechanisms of these diseases and aid in the development of potential therapeutic targets.
