Parkin Antibody (PRK8) is a mouse monoclonal IgG2b kappa light chain antibody that targets Parkin, also known as PRKN, PARK2, or parkin RBR E3 ubiquitin protein ligase, with confirmed reactivity in human, mouse, and rat samples. Parkin is a 465-amino acid E3 ubiquitin ligase encoded by the PARK2 gene, essential for the ubiquitination process, which tags proteins for degradation, maintaining cellular homeostasis by eliminating damaged or misfolded proteins. Parkin is particularly significant in mitophagy, the selective autophagic removal of dysfunctional mitochondria, which is crucial for preserving mitochondrial quality and function. Mutations in the PARK2 gene are associated with autosomal recessive juvenile Parkinson′s disease, highlighting Parkin′s importance in neuronal health. Anti-Parkin antibody (PRK8) is raised against recombinant human Parkin, targeting an epitope within the protein′s C-terminal region. Parkin (PRK8) monoclonal antibody is validated for diverse applications, including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Parkin expression is observed in neurons across various brain regions, including the basal ganglia, midbrain, cerebellum, and cerebral cortex, with roles that are evolutionarily conserved across species. Anti-Parkin antibody (PRK8) serves as a critical tool for studying Parkin′s functions and involvement in cellular processes and neurodegenerative research.

What % of milk to you recommend? I get a lot of background in 1% milk and it is hard to tell where the band is. Is it ok to boil samples? Thank you!

I am trying to find out some good IF images in a reputed journal. Although in the reference you have mentioned in 2 Sci Advance journal. Unfortunately, I haven't found any IF-image in those papers. Do you have some good reference images?

Hello. I have tried the parkin antibody at 1:50, 1:100, 1:200 dilutions for immunofluorescent staining using retinal cryosections and agar-embedded vibratome slices with no detectable signal. Do you have any recommendations for troubleshooting? Thanks.

Hi. I am using this antibody to detect parkin in my cells (SH-SY5Y cells) via ICC. However, I am not able to detect any signal (I have used up to 1:50 dilution and have used both milk and bsa). Could you possibly give me an optimal protocol for ICC?

Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. SC-32282 is currently available conjugated to FITC, AF488, and AF647.
