Parkin Antibody (D-1) is a mouse monoclonal IgG2b kappa light chain antibody designed to target human Parkin, also known as PRKN or parkin RBR E3 ubiquitin protein ligase. Parkin is a 465-amino acid E3 ubiquitin ligase encoded by the PARK2 gene, essential for the ubiquitin-proteasome system by marking damaged or misfolded proteins for degradation, thereby preserving cellular protein homeostasis. Additionally, Parkin plays a critical role in mitophagy, the selective autophagic removal of dysfunctional mitochondria, which is vital for maintaining cellular energy balance and preventing oxidative stress. Structurally, Parkin contains a RING-between-RING (RBR) domain that facilitates its E3 ligase activity, enabling the transfer of ubiquitin from E2 enzymes to specific substrate proteins. Post-translational modifications such as phosphorylation and ubiquitination are crucial for regulating Parkin′s activity and localization within the cell, particularly movement from the cytoplasm to the outer mitochondrial membrane in response to mitochondrial damage. Within the cell, Parkin is primarily localized in the cytoplasm under normal conditions but translocates to damaged mitochondria to initiate mitophagy, interacting with proteins like PINK1, DJ-1, LRRK2, and alpha-synuclein, which are also implicated in Parkinson′s disease. These interactions underscore Parkin′s significance in neuronal health and involvement in the pathology of neurodegenerative diseases. Anti-Parkin antibody (D-1) was raised against amino acids 61-360 of human Parkin, mapping to an internal region of the protein, and exhibits reactivity in mouse, rat, and human samples.
