Parkin Antibody (D-1) is a mouse monoclonal IgG2b kappa light chain antibody designed to target human Parkin, also known as PRKN or parkin RBR E3 ubiquitin protein ligase. Parkin is a 465-amino acid E3 ubiquitin ligase encoded by the PARK2 gene, essential for the ubiquitin-proteasome system by marking damaged or misfolded proteins for degradation, thereby preserving cellular protein homeostasis. Additionally, Parkin plays a critical role in mitophagy, the selective autophagic removal of dysfunctional mitochondria, which is vital for maintaining cellular energy balance and preventing oxidative stress. Structurally, Parkin contains a RING-between-RING (RBR) domain that facilitates its E3 ligase activity, enabling the transfer of ubiquitin from E2 enzymes to specific substrate proteins. Post-translational modifications such as phosphorylation and ubiquitination are crucial for regulating Parkin′s activity and localization within the cell, particularly movement from the cytoplasm to the outer mitochondrial membrane in response to mitochondrial damage.
