PAI-1 Antibody (C-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects PAI-1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PAI-1 Antibody (C-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PAI-1, also known as SERPINE1, plays a crucial role in regulating fibrinolysis and tissue remodeling by inhibiting urokinase-type plasminogen activator (uPA) and tissue plasminogen activator (tPA), which are essential for the conversion of plasminogen to plasmin. This regulation is vital for maintaining hemostasis and preventing excessive tissue degradation, making PAI-1 a significant factor in various physiological and pathological processes, including wound healing, cancer progression, and cardiovascular diseases. PAI-1 can exist in both active and latent forms, with the latent form being able to regain activity through denaturation and renaturation, highlighting PAI-1′s dynamic role in the regulation of proteolytic activity. The ability of PAI-1 to fold into an inactive state without cleavage underscores the complexity of PAI-1′s function and importance in cellular processes.

Can the sc-5297 antibody be used for western blot of rat kidney tissue? I was trying to find PAI-1 antibody (sc-8979) and just find it has been replaced by sc-5297.

Hello, I have been using PAI-1 Antibody (C-9): sc-5297 for WB and IF successfully on human pulmonary artery endothelial cells. I am hoping to do similar experiments on bovine aortic endothelial cells--does this ab cross-react with bovine cells?

Thank you for your question. We have not tested this mouse monoclonal, PAI-1 Antibody (C-9): sc-5297, on samples of bovine origin to confirm reactivity with that species. The antibody is raised against a recombinant protein corresponding to amino acids 24-158 of PAI-1 of human origin. This sequence shares 83% sequence identity with bovine PAI-1 protein. Based on this alignment, there is a significant chance that the antibody would react with bovine. However, we can not recommend it for this species since we have not confirmed it experimentally.

Conditions: 12% gel electrophoresis, wet transfer over 2 hours, primary antibodies at 1:200 dilution in 5% mild blocking solution (4C overnight). There was no band visible following the protocol, although positive control (b-actin) was seen.
