p-MAPKAPK-2 Antibody (83.Thr 334) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Thr 334 phosphorylated MAPKAPK-2 protein of human origin by western blotting (WB) and enzyme-linked immunosorbent assay (ELISA). anti-p-MAPKAPK-2 antibody (83.Thr 334) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The p38 mitogen-activated protein kinase (MAPK) pathway plays a crucial role in mediating cellular responses to environmental stress, and MAPKAP kinases, including MAPKAPK-2, are key substrates in this signaling cascade. Upon activation, p38 phosphorylates MAPKAPK-2, which in turn phosphorylates several important nuclear targets such as CREB/ATF1 and serum response factor, as well as cytoplasmic targets like HSP25/27 and glycogen synthase. Phosphorylation of threonine 334, strategically located between the kinase domain and C-terminal regulatory domain, acts as a regulatory switch for nuclear import and export of MAPKAPK-2. This modification influences protein localization and functional interactions, as phosphorylated MAPKAPK-2 can mask nuclear localization signal while exposing nuclear export signal, facilitating p38 transport to cytoplasm. Understanding these phosphorylation dynamics is essential for elucidating broader implications of MAPKAPK-2 in stress response and cellular signaling pathways.
