HSP 27 Antibody (F-4) is a high quality monoclonal HSP 27 antibody (also designated HSPB1 antibody, heat shock protein family B (small) member 1 antibody, HSP27 antibody) suitable for the detection of the HSP27 (HSPB1) protein of mouse, rat and human origin. HSP 27 Antibody (F-4) is available as both the non-conjugated anti-HSP 27 antibody form, as well as multiple conjugated forms of anti-HSP 27 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The heat shock proteins (HSPs) comprise a group of highly conserved, abundantly expressed proteins with diverse functions, including the assembly and sequestering of multiprotein complexes, transportation of nascent poly-peptide chains across cellular membranes and regulation of protein folding. Heat shock proteins (also known as molecular chaperones) fall into six general families: HSP 90, HSP 70, HSP 60, the low molecular weight HSPs, the immunophilins and the HSP 110 family. The low molecular weight family includes HSP 10, HSP 20, HSP 27, HSP 32 and HSP 40. HSP 27 is a constitutively expressed cytoplasmic protein that co-localizes to the nucleus upon stress induced by insult. Heat, cytokines and hormones are among the factors that stimulate the synthesis of HSP 27. In vitro, HSP 27 becomes highly phosphorylated following exposure to stress. The discovery that HSP 27 is regulated by hormones such as estrogen has led to studies establishing a relationship between HSP 27 and breast cancer.

recommended for WBWorks very well for whole cell lysates of HEp-2 and RKO cells when the antibody is diluted 1:500 in BSA/PBS-T. For IF (cells were fixed in 2% formaldehyde) we had to dilute the antibody 1:50 to achieve a good cytoplasmic staining.

Detecting mouse HSP27 protein by WB using sc-13132Western blotting conditions:
Species: Mouse (C57BL6)
Tissue: Skeletal muscles
Protein amount: 10ug
SDS-PAGE gel%: 4-20% gradient gel or 16% gel
Blocking buffer: 5% skim milk in PBS
Primary antibody dilution: 1:1000 (1:500 - 1:2000 works well) in 5% milk-PBS
Washing buffer: TBS - TBST - TBS
Secondary antibody detection methods: HRP-conjugated in 5% milk-PBS
Band detected: below the 30 kDa protein marker, corresponding to the 27 kDa size of this protein
Film and Li-cor exposure: 5s and 15s, respectively
Precautions: HSP27 is quite abundantly expressed in murine skeletal muscles and sc-13132 detects this protein very well. If you load a very high amount of protein and use a high primary antibody amount, upon using the HRP-substrate you might get a 'burn-through effect' due to the signal generated and bands might begin to appear hollow.
Overall, the blot quality generated using this antibody and the above mentioned conditions was excellent!
