    HCN3 Antibody (A-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects HCN3 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-HCN3 antibody (A-3) is available as the non-conjugated form. Hyperpolarization-activated, cyclic nucleotide-binding channels (HCN) are essential for regulating neuronal excitability and cardiac rhythm, and are activated by the binding of intracellular cyclic nucleotides, which is crucial for their function. The HCN family, comprising four members (HCN1-4), features a core transmembrane segment domain along with a C-terminal cyclic nucleotide-binding domain that is 120 amino acids long. HCN channels are predominantly expressed in the brain, heart, thalamus, and testis, where these channels play a vital role in generating spontaneous rhythmic activity. Notably, HCN3 contains a unique segment characterized by a series of positively charged amino acids, known as the S4 region, which functions as the voltage sensor. In the central nervous system, HCN3 and HCN4 are primarily localized in the hypothalamus and thalamus, respectively, highlighting their importance in modulating various physiological processes.

    We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.
