H-Ras Antibody (M3) is a mouse monoclonal IgG1 kappa light chain antibody raised against recombinant human H-Ras, a crucial small GTPase involved in cellular signal transduction. Anti-H-Ras antibody (M3) recognizes an epitope within the H-Ras immunogen sequence and demonstrates broad species cross-reactivity, detecting H-Ras in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry on paraffin-embedded sections (IHCP) applications. As a member of the Ras gene family alongside K-Ras and N-Ras, H-Ras (also known as Harvey rat sarcoma viral oncogene homolog) undergoes sophisticated regulation through its GTPase cycle, switching between inactive GDP-bound and active GTP-bound states. The functionality of H-Ras depends on critical post-translational modifications at the C-terminal CAAX motif: farnesylation adds a farnesyl lipid group to the cysteine residue enabling membrane anchoring, palmitoylation attaches palmitoyl groups directing H-Ras to specific membrane microdomains, and methylation stabilizes these membrane interactions. These modifications prove essential for H-Ras′s proper subcellular localization and ability to interact with both upstream activators and downstream effectors. When these regulatory mechanisms become disrupted, the resulting aberrant signaling can promote oncogenic transformation and tumor development. Through these diverse cellular functions, H-Ras (M3) antibody provides researchers with a valuable tool for investigating Ras-mediated signaling pathways and cancer biology mechanisms.
