GADD 34 Antibody (B-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects GADD 34 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-GADD 34 antibody (B-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GADD 34, also known as PPP1R15A, plays a crucial role in cellular response to stress, particularly in DNA damage context. GADD 34 functions as a regulatory protein that interacts with protein phosphatase 1 (PP1), facilitating dephosphorylation of eukaryotic translation initiation factor 2 alpha (eIF2α). This action is vital because eIF2α phosphorylation state determines protein synthesis during stress conditions; when eIF2α is phosphorylated, global protein synthesis reduces, allowing cells to conserve resources and focus on repairing damage. GADD 34′s ability to modulate this pathway underscores its importance in maintaining cellular homeostasis and preventing damaged DNA propagation, which can lead to tumorigenesis. GADD 34 also interacts with other proteins involved in stress responses, highlighting its role in cellular signaling networks.

Rated 1 out of 5 by from Antibody not working as expectedI have tried this antobody in a dilution 1:1000 in TBST overnight for HaCat immortalized cell lysates multiple times and and no band appears when revealing the membrane I have used Tunycamicin and DTT as positive controls but no results. If anybody has suggestions I would greatly appreciate it.

Rated 1 out of 5 by from Garbage antibodyRepeated attempts to validate this antibody (at 1:500 in PBST w/ 5% milk and 2 mM azide) from lysates of HEK293T and HeLa cells subjected to a 5 hr treatment with 0.4 uM thapsigargin (as in Novoa et al., 2001, JCB) failed to elicit a signal. Plausible that this antibody just came from a bad lot (#D1719), but I'm not wasting any more of my time attempting to coax it into functionality.

Rated 4 out of 5 by from Works well in human Hap-1 cellsI used the GADD34 antibody in Hap-I cells (total input 50mcg) in TBS-T and blocked with 5% milk and got a specific band. Protein input has to be quite high to see a band.
