GADD 153 Antibody (9C8) is a mouse monoclonal IgG2b kappa light chain antibody designed to detect GADD 153 protein, also known as CHOP or DDIT3, across mouse, rat, and human species using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP) techniques. GADD 153 monoclonal antibody (9C8) targets an epitope derived from the mouse origin of GADD 153 and is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants, to suit diverse experimental needs. GADD 153 is a crucial C/EBP-related nuclear transcription factor localized within the cell nucleus, where GADD 153 plays a pivotal role in regulating gene expression in response to cellular stressors such as nutrient deprivation and metabolic disturbances. By residing in the nucleus, GADD 153 can effectively interact with other transcription factors, facilitating its function in halting cell cycle progression from the G1 to S phase. GADD 153 achieves this by forming dimers with other C/EBP proteins, thereby redirecting these complexes from classical C/EBP binding sites to unique nonclassical sites, acting as a negative modulator of C/EBP-like proteins in terminally differentiated cells. This regulatory mechanism is analogous to how Id proteins modulate the activity of MyoD and related proteins during muscle cell development. Understanding the nuclear localization of GADD 153 is essential, as this localization underscores GADD 153′s role in orchestrating cellular responses to stress at the transcriptional level, making anti-GADD 153 antibody (9C8) an invaluable tool for researchers investigating cell cycle regulation, stress response pathways, and related molecular mechanisms.
