GADD 153 Antibody (6D313) is a mouse monoclonal IgG2b kappa light chain antibody engineered to detect GADD 153, also known as CHOP or DDIT3, across mouse, rat, and human species using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). GADD 153 monoclonal antibody (6D313) targets an epitope derived from mouse GADD153, ensuring precise and reliable binding for various experimental applications. GADD 153 (6D313) antibody recognizes a critical C/EBP-related nuclear transcription factor characterized by its basic leucine zipper (bZIP) domain, which is essential for dimerization with other C/EBP family members and for binding to specific DNA sequences to regulate gene expression. The structural integrity of the bZIP domain determines GADD 153′s ability to form homodimers or heterodimers, thereby influencing regulatory functions in response to cellular stressors such as nutrient deprivation and metabolic disturbances. This structural feature allows GADD 153 to undergo conformational changes that enable interaction with nonclassical DNA binding sites, effectively altering the transcriptional landscape to induce cell cycle arrest at the G1 to S phase transition and promote apoptotic pathways. Additionally, the dimerization capability of GADD 153 is crucial for its role as a negative modulator of C/EBP-like proteins in terminally differentiated cells, similar to how Id proteins regulate MyoD activity during muscle cell differentiation. These detailed structural characteristics underscore GADD 153′s significance in orchestrating cellular responses to stress at the transcriptional level, making anti-GADD 153 antibody (6D313) an invaluable tool for researchers investigating molecular mechanisms underlying cell cycle regulation, apoptosis, and stress-induced transcriptional modulation.

Can GADD 153 (6D313): sc-71136 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Professor Griffin
Thank you for your question. Yes, GADD 153 (6D313): sc-71136 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
