GAD-65 Antibody (GAD-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects GAD-65 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-GAD-65 antibody (GAD-6) specifically recognizes the GAD-65 isoform, which plays a crucial role in the synthesis of gamma-aminobutyric acid (GABA), the primary inhibitory neurotransmitter in the central nervous system. GAD-65 (GAD-6) monoclonal antibody recognizes a protein predominantly localized to synaptic vesicle membranes in nerve terminals, where GAD-65 is essential for the rapid production of GABA in response to increased neuronal activity. GAD-65 ability to respond to heightened demands for GABA is vital for maintaining proper neurotransmission and preventing excitotoxicity. In contrast, GAD-67, which is also present in the brain, is responsible for maintaining basal GABA levels and is distributed throughout the cell. The interplay between these two isoforms is critical; while GAD-65 loss can lead to impaired GABA neurotransmission, GAD-67 absence is associated with severe consequences.
