GAD-65/67 Antibody (C-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects GAD-65/67 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GAD-65/67 (C-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The GAD-65 and GAD-67 proteins, which are crucial for the synthesis of gamma-aminobutyric acid (GABA) from L-glutamic acid, play a significant role in neurotransmission within the brain. GAD-65, localized to synaptic vesicle membranes in nerve terminals, is activated during increased demand for GABA, while GAD-67, which is distributed throughout the cell, maintains basal GABA levels essential for normal neuronal function. The proper functioning of these proteins is vital, as the loss of GAD-65 can disrupt GABA neurotransmission, leading to neurological issues, whereas the absence of GAD-67 is lethal, underscoring the importance of these proteins in maintaining the balance of inhibitory signaling in the brain. Additionally, GAD-67 has two isoforms due to alternative splicing, with the predominant form being GAD-67 and a minor form known as GAD-25, which is not expressed in the brain but is found in various endocrine tissues.
