ERK 1 Antibody (E-6) is a mouse monoclonal IgG1 kappa light chain antibody specific for an epitope mapping between amino acids 291-335 within subdomain XI of ERK 1 of rat origin, exhibiting strong species reactivity with mouse, rat, and human proteins by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA); anti-ERK1 antibody (E-6) is also reactive with additional species including equine, canine, bovine, and porcine. ERK1, also known as Mitogen-activated protein kinase 3 (MAPK3), is a crucial component of the MAPK/ERK signaling pathway, which transmits signals from the cell surface to the nucleus in response to various extracellular stimuli such as growth factors, hormones, and cytokines. ERK1 interacts with a variety of proteins that regulate activation and function. Upstream, ERK1 is activated by dual phosphorylation by MAP kinase kinases MEK1 and MEK2, which themselves are activated by Raf kinases like Raf-1. ERK1 interacts with scaffold proteins such as KSR1 and MP1, which facilitate the assembly of the MAPK signaling module, ensuring specificity and efficiency of signal transduction. Once activated, ERK1 translocates to the nucleus where ERK1 interacts with and phosphorylates numerous transcription factors, including ELK1, c-Fos, and c-Jun, leading to the regulation of genes involved in cell proliferation, differentiation, and survival. ERK1 also interacts with downstream kinases such as RSK (ribosomal S6 kinase), which phosphorylate additional substrates involved in protein synthesis and cell growth. ERK1 interactions with these proteins are essential for the propagation and modulation of cellular responses to external stimuli, and dysregulation of ERK1 signaling is implicated in various diseases, including cancer and neurodegenerative disorders. ERK1 monoclonal antibody (E-6) is an invaluable tool for researchers studying ERK1 interactions and the role of the MAPK/ERK pathway in health and disease.
