DOHH Antibody (E-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects DOHH in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DOHH (E-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DOHH, or deoxyhypusine hydroxylase/monooxygenase, also referred to as HLRC1 (HEAT-like (PBS lyase) repeat containing 1), is a metalloenzyme that plays a crucial role in the synthesis of hypusine, a unique amino acid modification essential for the function of eukaryotic translation initiation factor 5A (eIF5A). DOHH is characterized by eight tandem HEAT repeats, which are vital for structural integrity and enzymatic activity. DOHH catalyzes the hydroxylation of a deoxyhypusine-containing intermediate formed by deoxyhypusine synthase, thereby converting it into hypusine and activating eIF5A, which is necessary for cell proliferation and growth. The evolutionary conservation of DOHH, along with interaction with eIF5A and deoxyhypusine synthase, underscores importance in cellular processes. Notably, inhibition of DOHH has been linked to cell cycle arrest at the G1/S boundary, suggesting potential as a target for antitumor therapies.
