CD71 Antibody (G-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD71 protein from mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CD71 (G-8) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD71, also known as transferrin receptor (TFR), is a type II membrane glycoprotein forming a disulfide-linked homodimer of two identical subunits. CD71 maintains iron homeostasis by binding to transferrin, a serum iron-transport protein, and facilitates cellular iron uptake through receptor-mediated endocytosis. CD71 expression remains high in proliferating cells, reticulocytes, and erythroid precursors, while staying absent in resting leukocytes but becoming upregulated upon activation of lymphocytes, monocytes, and macrophages. CD71 appears on most dividing cells and brain endothelium, showing significance in hematopoiesis and neurobiology. A second transferrin receptor, TFR2, mediates transferrin-bound iron uptake and appears predominantly in liver, hepatocytes, and erythroid precursors. TFR2 gene mutations lead to hereditary hemochromatosis type III (HFE3), an iron overload disorder primarily affecting Caucasians, highlighting CD71′s importance in iron metabolism and related pathologies.
