CD71, also known as transferrin receptor 1 (TfR1), is a transmembrane glycoprotein that plays a critical role in cellular iron uptake. Its primary function is to bind transferrin, the main carrier protein for iron in the blood, and facilitate the internalization of transferrin-bound iron into cells through receptor-mediated endocytosis. Iron is an essential nutrient required for numerous cellular processes, including DNA synthesis, energy production, and oxygen transport. CD71 is highly expressed on the surface of various cell types, particularly those with high proliferative rates such as rapidly dividing cells in the bone marrow. Activation of CD71 involves several steps aimed at promoting its function in iron uptake. Upon binding to transferrin, CD71 undergoes conformational changes that facilitate the formation of clathrin-coated pits on the cell membrane. These pits serve as entry points for the internalization of transferrin-bound iron into the cell through endocytosis. Following endocytosis, the transferrin-CD71 complex is trafficked to early endosomes, where the acidic pH triggers the release of iron from transferrin. Subsequently, the iron is transported across the endosomal membrane into the cytoplasm, where it can be utilized for various cellular processes. CD71-containing endosomes then undergo recycling back to the cell surface, where CD71 is made available for further rounds of iron uptake. Overall, the activation of CD71 is tightly regulated to ensure efficient cellular iron acquisition and maintenance of iron homeostasis.
