Caspase-8 p10 Antibody (C502S) is a mouse monoclonal IgG1 antibody engineered against a synthetic peptide corresponding to the N-terminus of the human caspase-8 p10 subunit. Anti-caspase-8 p10 antibody (C502S) effectively recognizes human caspase-8 in western blotting (WB) and immunoprecipitation (IP) assays without cross-reacting with its precursor forms, ensuring precise detection of the active enzyme. Caspase-8 is a pivotal initiator caspase located primarily in the cytoplasm, where caspase-8 plays a critical role in the extrinsic apoptosis pathway. Upon activation of death receptors such as TNFRSF1A and TNFRSF6/FAS, caspase-8 is recruited to the death-inducing signaling complex (DISC) through interaction with the adaptor molecule FADD. This localization to the DISC facilitates rapid activation of caspase-8, which subsequently cleaves and activates downstream effector caspases, leading to the execution of apoptosis. Cytoplasmic presence of caspase-8 is essential for swift response to apoptotic signals, thereby maintaining cellular homeostasis and immune function. Caspase-8 is ubiquitously expressed with particularly high levels in peripheral blood leukocytes, thymus, liver, and spleen. Mutations or deficiencies in the CASP8 gene, which encodes caspase-8, can result in CASP8 deficiency disorder (CASP8D), characterized by splenomegaly and impaired apoptosis of peripheral blood leukocytes, ultimately leading to immunodeficiency due to defects in T lymphocyte, NK cell, and B lymphocyte activation. Caspase-8 p10 (C502S) monoclonal antibody is an essential tool for researchers studying apoptosis, immune regulation, and related pathologies, providing reliable detection of active caspase-8 within critical cellular context.
