Caspase-3 p17 Antibody (G-5) is a mouse monoclonal IgG2b kappa light chain antibody designed to detect the p17 subunit of caspase-3, also known as apopain, SCA-1, Yama, and CPP32. Anti-caspase-3 antibody (G-5) targets amino acids 56-104 near the N-terminus of human caspase-3, enabling reliable detection of both the p17 subunit and full-length procaspase-3 across mouse, rat, and human species through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Caspase-3 is a crucial aspartate-specific cysteine protease located in the cytoplasm of cells, where caspase-3 plays an essential role in the execution phase of apoptosis, the programmed cell death process vital for development, tissue homeostasis, and elimination of damaged or unwanted cells. The cytoplasmic localization of caspase-3 allows quick interaction with and cleavage of various substrates upon activation, facilitating orderly dismantling of cellular components during apoptosis. During this process, procaspase-3 is cleaved to form the active p17 subunit, which dimerizes with the p11 subunit to create an active enzyme capable of targeting key proteins like PARP, thereby driving the apoptotic cascade forward. Caspase-3 activates downstream caspases such as caspase-6, -7, and -9, amplifying the apoptotic signal and ensuring thorough execution of cell death. The strategic cytoplasmic location of caspase-3 is crucial for rapid response to apoptotic signals, making caspase-3 an indispensable component in various physiological processes and disease pathways, including cancer, neurodegeneration, and immune responses. With high expression levels in vital organs such as lung, spleen, heart, liver, kidney, and immune cells, caspase-3′s widespread presence underscores a fundamental role in maintaining cellular integrity and organismal health. By providing robust and reliable detection of caspase-3 in multiple experimental settings, caspase-3 p17 (G-5) monoclonal antibody is an essential tool for researchers investigating molecular mechanisms of apoptosis and related cellular processes.

Can caspase-3 p17 (G-5): sc-166589 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Trav11
Thank you for your question. Yes, caspase-3 p17 (G-5): sc-166589 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
