caspase-3 Antibody (7.1.44) is a mouse monoclonal IgG2a kappa light chain antibody engineered to detect the full-length human caspase-3 protein, also known as apopain, SCA-1, Yama, and CPP32. Anti-caspase-3 antibody (7.1.44) recognizes caspase-3 across multiple species, including mouse, rat, and human, and is recommended for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Caspase-3 is a crucial aspartate-specific cysteine protease within the ICE subfamily, primarily located in the cytoplasm of cells where caspase-3 plays a pivotal role in the execution phase of apoptosis. During apoptosis, caspase-3 is activated from its inactive precursor form through proteolytic cleavage, forming an active heterodimer composed of p17 and p11 subunits. This active enzyme orchestrates the systematic dismantling of cellular components by cleaving key substrates such as PARP, thereby ensuring orderly cell death. Additionally, caspase-3 facilitates the release of SREBPs from the endoplasmic reticulum membrane and activates other caspases like caspase-6, -7, and -9, amplifying the apoptotic signal. The precise cytoplasmic localization of caspase-3 is essential for maintaining cellular homeostasis by eliminating damaged or unnecessary cells, which is vital for processes such as development, immune response, and cancer prevention. Caspase-3 (7.1.44) antibody is available in both non-conjugated form and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for diverse research needs. Caspase-3 (7.1.44) monoclonal antibody is an indispensable tool for scientists investigating apoptosis and related cellular mechanisms, making caspase-3 essential for research areas including cancer biology, immunology, and neurodegenerative diseases.

How much sample do you recommend loading on the gel for Western blot detection using caspase-3 (7.1.44): sc-56051?

Asked by: cjMara
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
