Caspase-3 p11 Antibody (C-6) is a mouse monoclonal IgG1 lambda light chain antibody designed to detect the p11 subunit of caspase-3, also known as apopain, SCA-1, Yama, and CPP32. Anti-caspase-3 p11 antibody (C-6) targets an epitope between amino acids 185-211 at the N-terminus of human caspase-3 p11, ensuring accurate recognition of both the p11 subunit and the full-length procaspase-3 in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) assays. Caspase-3 is a vital aspartate-specific cysteine protease within the ICE subfamily, playing a crucial role in the execution phase of apoptosis, the programmed cell death essential for development, tissue homeostasis, and eliminating damaged or unwanted cells. Structurally, caspase-3 is synthesized as an inactive proenzyme, procaspase-3, which undergoes post-translational proteolytic cleavage at specific aspartate residues to form the active enzyme composed of two p17 and two p11 subunits, creating a heterotetramer necessary for enzymatic activity. These post-translational modifications enable caspase-3 to cleave key substrates such as PARP and facilitate the orderly disassembly of cellular components during apoptosis. Active caspase-3 can activate downstream caspases like caspase-6, -7, and -9, thereby amplifying the apoptotic signal and ensuring thorough execution of cell death. Caspase-3 is predominantly localized in the cytoplasm, allowing rapid response to apoptotic signals and interaction with various substrates to drive the cell death process. Caspase-3 is highly expressed in vital organs including the lung, spleen, heart, liver, kidney, and immune cells, underscoring its pivotal role in numerous physiological processes and disease mechanisms such as cancer, neurodegenerative disorders, and immune responses.
