c-Myc Antibody (9E11) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 408-420 of human c-Myc. c-Myc mouse monoclonal antibody (9E11) recognizes an epitope within this region and detects c-Myc-tagged fusion proteins from mouse, rat, and human sources. Anti-c-Myc monoclonal antibody (9E11) demonstrates high suitability for Western blotting, immunoprecipitation, immunofluorescence, and immunohistochemistry on paraffin-embedded tissues. c-Myc (9E11) antibody is available in non-conjugated form and multiple conjugated forms including agarose, HRP, PE, FITC, and various Alexa Fluor® conjugates, offering versatility for diverse experimental needs. The c-Myc protein, encoded by the MYC proto-oncogene on chromosome 8q24, is a nuclear transcription factor that plays a crucial role in regulating cell proliferation, differentiation, and apoptosis. c-Myc contains essential structural motifs in the C-terminal region, such as the basic region, helix-loop-helix, and leucine zipper domains, which are vital for DNA binding and dimerization with Max protein. The c-Myc/Max heterodimer binds to specific E-box sequences (CACGTG) in target gene promoters, activating transcription and promoting cell cycle progression. Overexpression or amplification of the c-Myc gene is a common feature in various human cancers, including Burkitt′s lymphoma, neuroblastoma, and carcinomas of the lung, breast, and colon, leading to uncontrolled cell growth and tumor development. Max can form heterodimers with other basic helix-loop-helix leucine zipper proteins like Mad1 and Mxi1; these complexes can repress transcription by recruiting co-repressors such as mSin3, thereby counteracting c-Myc activity. Understanding the complex regulatory mechanisms involving c-Myc is essential for cancer research and the development of targeted therapies.

What is the band size expected in western blotting when using 9E11 c-Myc antibody?

When should the TransCruz reagent sc-47694 X be used instead of the standard form sc-47694?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with c-Myc (9E11): sc-47694?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. Rated 5 out of 5 by from Great signalrimary antibody c-Myc (9E11) sc-47694 was used diluted 1:1000 according to the manufacturer's recommendations (for Western Blotting dilution range 1:100-1:1000), in Milk 5%. Membranes of PVDF have been incubated overnight at 4°C and then they have been probed for 1h at RT with secondary antibodies diluted 1:5000 in Milk 5%. We have tested this antibodiy three tumor cell lines: HeLa, A375 and glioblostoma primary cell line. In all the samples the recognition of the TAG was effective. The signal was clear and good, so we can conclude that the antibody has worked well. very well.
