eIF1AY Activadores

eIF1AY activators primarily influence the protein indirectly, based on its pivotal role in the initiation phase of eukaryotic translation. This protein enhances the fidelity of initiation by augmenting the binding of the initiator tRNA to the 40S ribosomal subunit in the P-site. While direct chemical activators for eIF1AY have not been widely recognized, there are several chemicals known to modulate the translation initiation process, thereby indirectly affecting eIF1AY's function.

Rapamycin and Torin 1, for instance, are inhibitors of mTOR, a critical protein for protein synthesis. By inhibiting the mTOR pathway, these chemicals can influence the translation initiation process, including the role of eIF1AY. Similarly, 4EGI-1, by inhibiting the eIF4E/eIF4G interaction, affects translation initiation dynamics that indirectly involve eIF1AY. Compounds like ISRIB enhance eIF2B activity under certain conditions, offering another avenue by which translation initiation can be modulated, thus potentially affecting eIF1AY's function. Salubrinal, which inhibits eIF2α dephosphorylation, and Guanabenz, which impacts eIF2α phosphorylation, also fall within this realm of influencing eIF1AY by modulating other components of the translation initiation process. Furthermore, chemicals like Perifosine, LY294002, and Wortmannin act on the Akt/mTOR and PI3K pathways, respectively. These pathways play substantial roles in translation, and by influencing them, these chemicals indirectly activate eIF1AY. Lastly, Resveratrol, known to influence protein synthesis via the mTOR pathway, provides another example of an indirect activator.
