Ionomycin functions by increasing intracellular calcium, which activates calcium/calmodulin-dependent protein kinases known to phosphorylate Zfp931, resulting in its activation. Similarly, thapsigargin acts by inhibiting the calcium ATPase of the sarco/endoplasmic reticulum, causing an increase in cytosolic calcium levels, which then activates kinases capable of phosphorylating Zfp931.
Further along these lines, calyculin A and okadaic acid prevent protein phosphatases 1 and 2A from dephosphorylating Zfp931, thereby keeping Zfp931 in an active state. Anisomycin activates stress-activated protein kinases, which are known to phosphorylate Zfp931, resulting in its activation. Dibutyryl-cyclic AMP (db-cAMP), a synthetic analog of cAMP, activates PKA, which subsequently phosphorylates and activates Zfp931. Epidermal Growth Factor (EGF) binds to its receptor and initiates a cascade of kinase activations, including those that can phosphorylate Zfp931. Bisindolylmaleimide I, at sub-activating concentrations for PKC, can paradoxically activate PKC, which then phosphorylates Zfp931. Retinoic acid is involved in various differentiation pathways and can promote the activation of kinases that phosphorylate Zfp931. Lastly, zinc sulfate provides zinc ions that serve as essential cofactors for various kinases; these kinases can phosphorylate Zfp931, which increases its activity. Each of these chemicals engages specific cellular signaling pathways that converge on the phosphorylation and subsequent activation of Zfp931, underscoring the diverse mechanisms by which this protein can be regulated.
