USP26 Inhibitors

Chemical inhibitors of USP26 include a variety of compounds that impede its function through different mechanisms. E-64 acts by irreversibly inhibiting cysteine proteases, which are crucial for protein degradation and recycling within the cell. By targeting these proteases, E-64 can alter the ubiquitin processing capabilities within the cell, thereby potentially inhibiting the deubiquitination activity of USP26. Similarly, Leupeptin Hemisulfate, a reversible inhibitor, targets serine and cysteine proteases, and its action may affect the proteolytic processing of proteins interacting with USP26, leading to inhibition of USP26's functional role in the cell. MG-132 is a proteasome inhibitor that prevents the degradation of polyubiquitinated proteins. This accumulation can indirectly inhibit USP26's deubiquitinating activity due to reduced availability of substrates. Lactacystin, another proteasome inhibitor, can lead to feedback inhibition on USP26's activity through the buildup of ubiquitinated proteins that are not being degraded effectively. PI-1840, by inhibiting the threonine protease activity of the proteasome, also contributes to the disruption of the cellular protein degradation machinery, potentially limiting USP26's role in the process.

Further inhibiting the deubiquitination process, PR-619 broadly targets deubiquitinating enzymes and can bind directly to the active site of USP26, obstructing its ability to process ubiquitin chains. b-AP15 and VLX1570 similarly inhibit the deubiquitinating enzymes USP14 and UCHL5 at the proteasome, which may indirectly lead to the inhibition of USP26 by changing the dynamics of proteasome-associated deubiquitinating activity and thus affecting the ubiquitin-dependent signaling pathways where USP26 is active. WP1130 and HBX 41108 selectively inhibit other deubiquitinating enzymes and, through their action, can lead to the inhibition of USP26 by steric hindrance or allosteric effects upon binding to enzymes with similar functions. IU1, by inhibiting USP14, can induce a compensatory cellular response that may result in the indirect inhibition of USP26. Lastly, Ubiquitin Aldehyde can bind to the active site of many deubiquitinating enzymes, including USP26, preventing the cleavage of ubiquitin from substrate proteins and thus inhibiting USP26's deubiquitinating function.
