UGGT1 Inhibitors

UGGT1 inhibitors, in the broadest sense, include compounds that interfere with protein folding, glycosylation, and ER homeostasis. Tunicamycin, for instance, inhibits N-linked glycosylation, a process that UGGT1 uses to mark misfolded proteins for further processing. These compounds alter ER homeostasis in different ways, such as inhibiting the ER Ca2+ ATPase (Thapsigargin), increasing intracellular calcium concentration (Ionomycin and A23187), or interfering with glycolysis (2-Deoxy-D-Glucose). Each of these changes to the ER environment can potentially influence the function of UGGT1, which plays a crucial role in maintaining protein quality control under conditions of ER stress.

Lastly, agents like DTT, Eeyarestatin I, MG-132, Cycloheximide, Geldanamycin, and Azetidine-2-carboxylic acid can indirectly influence UGGT1 by affecting protein folding or degradation processes. DTT, for example, reduces disulfide bonds and can lead to protein misfolding, potentially increasing the number of substrates for UGGT1. Eeyarestatin I and MG-132 inhibit ER-associated degradation and proteasome activity, respectively, possibly leading to an accumulation of misfolded proteins that would be substrates for UGGT1. Cycloheximide inhibits protein synthesis, thereby reducing the number of proteins that could potentially misfold and become substrates for UGGT1. Geldanamycin, by binding to Hsp90, impacts protein folding pathways, potentially influencing UGGT1's role in addressing misfolded proteins. Azetidine-2-carboxylic acid can cause protein misfolding by being incorporated into proteins in place of proline, potentially affecting the activity of UGGT1.
