Phorbol 12-myristate 13-acetate (PMA) is another activator that directly engages protein kinase C (PKC), which phosphorylates and activates TTC24.
Beyond these, the inhibition of phosphatases by Okadaic Acid maintains TTC24 in a phosphorylated state, which is associated with its activity. This is because the inhibition of phosphatases prevents the dephosphorylation of TTC24, thus sustaining its activation. Anisomycin operates through a different mechanism, activating the JNK and p38 MAP kinase pathways, both of which are capable of phosphorylating TTC24. The natural compound Epigallocatechin gallate (EGCG) activates a complex network of signaling pathways, including kinases that may activate TTC24. Sphingosine can be metabolized into sphingosine-1-phosphate (S1P), which through its receptors can activate kinases that influence the phosphorylation and activation of TTC24. Compounds like H-89 and Bisindolylmaleimide I, although primarily known as kinase inhibitors, can have off-target effects that result in the activation of kinases which subsequently activate TTC24. Thapsigargin and Calyculin A disrupt normal phosphatase activities leading to increased calcium levels or inhibition of dephosphorylation activities, respectively, which can result in the activation of TTC24. Lastly, Dibutyryl-cAMP, a stable cAMP analog, directly activates PKA, which is known to phosphorylate and activate TTC24, ensuring that the protein is in an active state. Each of these chemicals engages specific biochemical pathways that converge on the activation of TTC24, ensuring its role in cellular signaling is maintained.
