TRS85 Activators

TRS85 is a protein that functions as a subunit of the TRAPP (Transport Protein Particle) complex, which is involved in vesicular transport processes within the cell. Specifically, TRS85 is a component of the TRAPP III complex, which has been implicated in autophagy, a cellular degradation and recycling process that is essential for cell survival and homeostasis. The TRAPP complexes, including TRAPP III where TRS85 is found, act as guanine nucleotide exchange factors (GEFs) for specific Rab GTPases, which are key regulators of intracellular membrane trafficking. By promoting the exchange of GDP for GTP on Rab proteins, TRAPP complexes facilitate the transition of Rabs into their active GTP-bound state, allowing them to recruit downstream effectors for vesicle formation, movement, and fusion.

TRS85, along with other TRAPP III-specific components, is believed to regulate autophagy by mediating the trafficking of autophagosomes to lysosomes. Autophagosomes are double-membraned vesicles that sequester cellular components for degradation, and their fusion with lysosomes is a critical step in the autophagic process. The precise mechanisms by which TRS85 and the TRAPP III complex contribute to this process are an active area of research. In yeast (Saccharomyces cerevisiae), TRS85 is known as a non-essential subunit that is specifically required for the TRAPP III complex function. In mammalian cells, the functional orthologs of TRS85 are thought to play similar roles in autophagy, although the composition and regulation of the TRAPP complexes may differ.
