TRMT61B Inhibitors

S-Adenosylhomocysteine, 5'-Methylthioadenosine, and 3-Deazaneplanocin A are examples of compounds that can inhibit methyltransferase activity by either directly interacting with the active site or by modulating levels of essential cofactors or substrates necessary for enzymatic activity. These molecules exert their inhibitory effect by either mimicking substrates or products of the methylation reaction, or by depleting the pool of cofactors required for the enzyme's action. Compounds such as Adenosine dialdehyde and Folic Acid Antagonists indirectly inhibit methyltransferases through the modulation of metabolic pathways that supply cofactors for methylation reactions. Nucleoside analogs like Decitabine and Azacitidine, while primarily used to affect DNA methylation, may also have an impact on RNA methylation processes. Hydralazine and RG108, although they are primarily associated with DNA methylation inhibition, might exert similar effects on RNA methyltransferases due to the similarities in methylation mechanisms. BIX-01294 and Chaetocin are inhibitors that target histone methyltransferases, but due to the conserved nature of the methylation domain, these inhibitors might also affect TRMT61B. It is important to note that the specificity and direct effects of these inhibitors on TRMT61B would require further experimental validation to confirm any actual inhibitory action.
