Zinc Chloride can directly enhance TRIM43's function by binding to its RING domain, crucial for its ubiquitin ligase activity, thereby catalyzing the conjugation of ubiquitin to substrate proteins. Similarly, Copper(II) sulfate can bind to TRIM43, potentially increasing its enzymatic activity by stabilizing the protein's structure. Small molecule activators like Sodium fluoride and Forskolin can activate kinases that phosphorylate TRIM43, with Sodium fluoride acting through the enhancement of kinase activity, while Forskolin elevates cAMP levels to activate protein kinase A (PKA). PKA then phosphorylates TRIM43, which increases its ubiquitin ligase activity.
Further, the influx of calcium ions into the cell via Ionomycin activates calcium-dependent kinases that can phosphorylate TRIM43, leading to its activation. Lastly, MG132, a proteasome inhibitor, can indirectly enhance the E3 ligase activity of TRIM43 by preventing the degradation of ubiquitinated proteins, thus potentially increasing the availability of substrates for TRIM43-mediated ubiquitination.
