Chemicals such as Forskolin and Phorbol 12-Myristate 13-Acetate (PMA) activate intracellular signaling cascades, like the cAMP pathway and PKC activation, respectively. By influencing these pathways, they can facilitate the activity of the Mediator complex, thereby indirectly affecting TRAP95.

Similarly, the chromatin landscape plays a crucial role in determining the Mediator complex's accessibility to DNA. Chemicals like Trichostatin A (TSA) function as histone deacetylase inhibitors, leading to a more open chromatin structure. This modification provides a conducive environment for the Mediator complex to function effectively. On the other hand, compounds like MG-132 and 5-Azacytidine exert their effects by modulating protein stability or altering the DNA methylation profile.
