Chemical activators of TPSB3 can initiate a cascade of intracellular events leading to its activation through several pathways, predominantly involving the increase of cyclic AMP (cAMP) levels. Forskolin directly stimulates adenylate cyclase, the enzyme responsible for converting ATP into cAMP. The elevated levels of cAMP can then activate protein kinase A (PKA), which may phosphorylate and activate TPSB3. Similar to forskolin, isoproterenol, a beta-adrenergic agonist, and salbutamol, a beta2-adrenergic agonist, both bind to their respective receptors and activate adenylate cyclase, consequently raising cAMP levels and activating PKA, which in turn activates TPSB3. Histamine, by stimulating H2 receptors, and adenosine, through interaction with A2 receptors, both G protein-coupled receptors, also activate adenylate cyclase, thus following the same pathway toward the activation of TPSB3.
Furthermore, dopamine, through D1-like receptors, and epinephrine, via beta-adrenergic receptors, both contribute to the activation of adenylate cyclase, following through the cAMP and PKA activation route, leading to the activation of TPSB3. Cholera toxin exerts a more sustained effect by catalyzing the ADP-ribosylation of the Gs alpha subunit, which in turn leads to continuous activation of adenylate cyclase and prolonged cAMP production, thereby ensuring prolonged activation of TPSB3 via PKA. Parathyroid hormone engages with its receptor to activate adenylate cyclase, while terbutaline, as a beta2-adrenergic agonist, functions similarly to salbutamol, enhancing cAMP levels and facilitating PKA-mediated activation of TPSB3. Lastly, vasoactive intestinal peptide, by interacting with VPAC receptors, triggers adenylate cyclase activity and thus increases cAMP, paving the way for PKA to activate TPSB3. Each of these chemicals, by increasing cAMP levels, ensures the activation of PKA, which then leads to the activation of TPSB3 through phosphorylation.
