TNFβ Inhibitors

The chemical class termed TNFβ inhibitors refers to a group of compounds that can interfere with the function of TNFβ, a cytokine involved in immune system regulation and inflammatory processes. The compounds listed operate through a variety of mechanisms, mainly targeting the NF-κB signaling pathway, which is crucial for the transcription of many inflammatory genes, including those related to the TNF family. Inhibition of this pathway results in a decreased expression of various cytokines and can dampen the inflammatory response. Chemicals such as thalidomide, BAY 11-7082, PDTC, parthenolide, curcumin, resveratrol, andrographolide, emodin, isoliquiritigenin, and sulforaphane exhibit their inhibitory effects by preventing the activation of NF-κB, which in turn can influence TNFβ's function.

Other compounds such as ruxolitinib and SP600125 target different components of the cytokine signaling pathways, namely JAK-STAT and JNK pathways, respectively. By altering these signaling pathways, the chemicals can modulate the activity of various cytokines, including TNFβ. These compounds are not exclusive TNFβ inhibitors; instead, they exhibit a broader range of activity across different signaling pathways that intersect with TNFβ's role in the immune response. The modulation of these pathways demonstrates the complex network of interactions that can be targeted to affect the function of a specific cytokine such as TNFβ.
