TGN38B Inhibitors

Chemical inhibitors of TGN38B can disrupt the protein's function by targeting various elements of the cellular machinery involved in protein trafficking and sorting. Manumycin A and FTI-277 work by selectively inhibiting enzymes like farnesyltransferase, a critical modifier of the Ras protein, which plays a significant role in signaling pathways that guide the localization and function of TGN38B. Similarly, GGTI-298 hampers the function of Rab proteins by preventing geranylgeranylation, a post-translational modification necessary for their activity. This disruption can indirectly affect the trafficking pathways that are crucial for the proper functioning of TGN38B. Meanwhile, SecinH3 targets cytohesins, disrupting ARF GTPase activity, which can lead to compromised vesicular trafficking and sorting, processes in which TGN38B is heavily involved.

Further affecting TGN38B's role in trafficking, ML141 acts as a Cdc42 inhibitor, impeding the regulation of vesicular trafficking, while compounds like Exo1 and Dyngo-4a inhibit Exo70 and dynamin, respectively, disrupting processes essential for vesicle tethering and release. CID-1067700 broadly disrupts the function of multiple GEFs for small GTPases, which are integral for the proper regulation of vesicular trafficking and protein sorting. This can lead to a chain of events culminating in the impairment of TGN38B's sorting functions. Additionally, Brefeldin A and Pitstop 2 interfere with the Golgi apparatus structure and clathrin-mediated endocytosis, respectively, both essential for the proper localization and function of TGN38B in protein sorting pathways. Lastly, BI-D1870's inhibition of RSK and Latrunculin B's disruption of actin polymerization can indirectly impair TGN38B by affecting the signaling pathways and cytoskeletal organization necessary for its function.

Manumycin A selectively inhibits Ras farnesyltransferase, an enzyme required for the proper localization and function of Ras proteins. TGN38B is involved in trafficking and sorting of proteins from the trans-Golgi network; inhibition of Ras can disrupt the trafficking pathways in which TGN38B functions.
