SUCO Inhibitors

SUCO Inhibitors have mechanisms that target diverse cellular components ranging from nuclear export proteins to elements of the cytoskeleton. For example, KPT-330 specifically inhibits XPO1, which is responsible for nuclear export, subsequently affecting SUCO's localization within the cell. Similarly, chemicals like Blebbistatin and Y-27632 act on the actomyosin contractile machinery. By inhibiting myosin II or ROCK, they influence the mechanical forces that stabilize the nuclear-cytoskeletal connections where SUCO acts as a linker. In this vein, inhibitors of microtubule dynamics such as Nocodazole and Taxol affect the LINC complex, to which SUCO is associated. These changes in microtubule stability consequently affect SUCO's functional state.

Another class of inhibitors like Trichostatin A and ALLN modify post-translational modifications protein degradation. Trichostatin A inhibits HDACs, leading to higher acetylation levels that affect the nuclear envelope structure, thereby disrupting SUCO's normal anchoring. ALLN inhibits calpain and proteasome activity, thus preserving the levels of proteins that interact with SUCO and can modify its functional state.
