SPLUNC3, or Short Palate, Lung, and Nasal Epithelium Clone 3, is a protein that belongs to the PLUNC (Palate, Lung, Nasal Epithelium Clone) family of proteins. It is encoded by the SPLUNC3 gene in humans. Members of this protein family are predominantly expressed in the upper airways and oral cavity and are thought to play a role in the innate immune defense against pathogens.
SPLUNC3 is secreted from epithelial cells and is found in various bodily fluids, including saliva and nasal secretions. The protein is characterized by its ability to interact with lipids and has been implicated in the regulation of surface liquid homeostasis in the respiratory tract. This property suggests a role for SPLUNC3 in maintaining the correct hydration of airway surfaces, which is essential for the proper functioning of the mucociliary clearance system-a defense mechanism that removes inhaled pathogens and debris from the respiratory tract.Moreover, SPLUNC3 has been shown to have bacteriostatic properties, meaning it can inhibit bacterial growth. This antibacterial activity is thought to stem from its ability to bind to bacterial lipopolysaccharides, thereby preventing the bacteria from adhering to and colonizing epithelial surfaces.Studies have also indicated that SPLUNC3 expression can be modulated in response to various environmental stimuli, including exposure to cigarette smoke, pathogens, and inflammatory cytokines, suggesting that it may have a role in the respiratory system's response to harmful insults.
