SERCA2 activators are chemicals that either directly or indirectly promote the function of the Sarco/Endoplasmic Reticulum Ca2+-ATPase 2 (SERCA2) protein. This protein plays an essential role in intracellular calcium homeostasis by pumping calcium ions from the cytosol back into the endoplasmic reticulum. Various chemical compounds can influence SERCA2 through different mechanisms. For example, Forskolin increases cAMP levels which subsequently activates PKA; this kinase phosphorylates SERCA2, enhancing its function. Caffeine, although commonly considered as an antagonist of calcium release channels, has the counterintuitive action of triggering SERCA2 to sequester elevated cytosolic calcium. Nitric oxide activates guanylate cyclase, which elevates cGMP levels. Elevated cGMP can indirectly stimulate SERCA2 activity. Thapsigargin and Ouabain, interestingly, induce compensatory mechanisms that eventually result in the upregulation of SERCA2.
Another group of compounds, such as Curcumin and Resveratrol, modulate intracellular calcium levels, triggering the activation of SERCA2 as a countermeasure to restore calcium homeostasis. Carbachol operates via Gq-protein signaling, triggering the mobilization of intracellular calcium and subsequently inducing SERCA2 activity to restore balance. Similarly, 2-APB alters intracellular calcium levels, which subsequently leads to SERCA2 activation. FTY720, known for its immunomodulatory effects, can deplete calcium from the endoplasmic reticulum, stimulating SERCA2 to pump calcium back in. Epicatechin and Sodium Nitroprusside are other nitric oxide elevators, like Nitric Oxide, which can indirectly activate SERCA2.
