Chemical inhibitors of SAMD1 can regulate its activity through various intracellular signaling pathways. Palbociclib, by inhibiting CDK4/6, can restrict the cell cycle's progression at the G1-S transition, a phase where SAMD1 is involved, thereby affecting its role in cell cycle regulation. Similarly, PD0325901 and U0126, both MEK inhibitors, can impede the MAPK/ERK pathway, which is crucial for SAMD1's function in cell differentiation and proliferation. This inhibition prevents the downstream signaling that SAMD1 participates in. SP600125 and SB203580 target different kinases within the MAPK pathway, JNK and p38 MAPK, respectively. The inhibition of these kinases can reduce SAMD1 activity related to stress response, apoptosis, inflammation, and cytokine production by disrupting the signaling cascades in which SAMD1 is active.
Further, LY294002 and Wortmannin, as PI3K inhibitors, can affect SAMD1 by blocking the PI3K/AKT pathway, consequently affecting processes such as cell survival and proliferation. Rapamycin, an mTOR inhibitor, can also disrupt the PI3K/AKT/mTOR pathway, thereby influencing SAMD1's role in cell metabolism and growth. Dasatinib's inhibition of Src family kinases can result in diminished SAMD1-related signal transduction and control of cell growth. Additionally, GW5074, a Raf kinase inhibitor, can disrupt the MAPK/ERK pathway upstream, thereby inhibiting the functional activity of SAMD1. Lastly, Y-27632 and ZM-447439, which are ROCK and Aurora kinase inhibitors, respectively, can alter SAMD1 activity; Y-27632 by affecting the organization of the actin cytoskeleton and ZM-447439 by interfering with chromosome alignment and segregation during mitosis, both processes where SAMD1 is implicated

|---|---|---|---|---|---|---|
Palbociclib | 571190-30-2 | sc-507366 | 50 mg | $321.00 | ||
Palbociclib is a CDK4/6 inhibitor, and as SAMD1 has been implicated in cell cycle regulation, this chemical can inhibit SAMD1 by hindering the cell cycle progression through G1-S phase, where SAMD1 is likely to be involved. | ||||||
SP600125 | 129-56-6 | sc-200635 sc-200635A | 10 mg 50 mg | $40.00 $150.00 | 257 | |
SP600125 is an inhibitor of JNK, which is part of the MAPK pathway. Given SAMD1's role in this pathway, inhibition of JNK can result in reduced activity of SAMD1 by preventing its involvement in stress response and apoptosis regulation. | ||||||
SB 203580 | 152121-47-6 | sc-3533 sc-3533A | 1 mg 5 mg | $90.00 $349.00 | 284 | |
SB203580 acts as a p38 MAPK inhibitor. By inhibiting p38 MAPK, SB203580 disrupts the pathway in which SAMD1 is active, particularly in inflammatory response and cytokine production, leading to the functional inhibition of SAMD1. | ||||||
LY 294002 | 154447-36-6 | sc-201426 sc-201426A | 5 mg 25 mg | $123.00 $400.00 | 148 | |
LY294002 is a PI3K inhibitor that can indirectly inhibit SAMD1 by blocking the PI3K/AKT pathway, which is involved in cell survival and proliferation, processes where SAMD1 is known to function. | ||||||
Rapamycin | 53123-88-9 | sc-3504 sc-3504A sc-3504B | 1 mg 5 mg 25 mg | $63.00 $158.00 $326.00 | 233 | |
Rapamycin inhibits mTOR, a critical component of the PI3K/AKT/mTOR pathway. Since SAMD1 has been associated with this pathway, rapamycin would inhibit SAMD1's role in processes like cell growth and metabolism. | ||||||
Wortmannin | 19545-26-7 | sc-3505 sc-3505A sc-3505B | 1 mg 5 mg 20 mg | $67.00 $223.00 $425.00 | 97 | |
Wortmannin is another PI3K inhibitor that can inhibit SAMD1 by blocking the PI3K/AKT pathway, thereby impeding cell survival and proliferation signals that SAMD1 may mediate. | ||||||
U-0126 | 109511-58-2 | sc-222395 sc-222395A | 1 mg 5 mg | $64.00 $246.00 | 136 | |
U0126 is an inhibitor of MEK1/2, which are upstream of ERK in the MAPK pathway. As SAMD1 is involved in this pathway, U0126 can inhibit SAMD1 by preventing the activation of ERK and subsequent signaling events related to cell differentiation that SAMD1 may influence. | ||||||
Dasatinib | 302962-49-8 | sc-358114 sc-358114A | 25 mg 1 g | $70.00 $145.00 | 51 | |
Dasatinib is a Src family kinase inhibitor. Src kinases are involved in multiple cell signaling pathways, including some that SAMD1 is part of. By inhibiting Src family kinases, dasatinib can inhibit SAMD1 activity related to signal transduction and control of cell growth. | ||||||
GW 5074 | 220904-83-6 | sc-200639 sc-200639A | 5 mg 25 mg | $106.00 $417.00 | 10 | |
GW5074 is a Raf kinase inhibitor, and as SAMD1 is involved in the MAPK/ERK signaling pathway, inhibiting Raf kinase would disrupt the pathway's function and therefore inhibit the functional activity of SAMD1. | ||||||
Y-27632, free base | 146986-50-7 | sc-3536 sc-3536A | 5 mg 50 mg | $186.00 $707.00 | 88 | |
Y-27632 is a ROCK inhibitor. The Rho/ROCK pathway plays a role in actin cytoskeleton organization, and SAMD1 is implicated in this pathway. Inhibition of ROCK would therefore inhibit SAMD1 by preventing the reorganization of the actin cytoskeleton that is essential for cell shape, motility, and possibly other functions where SAMD1 is involved. | ||||||
